چکیده

Please find attached the comments from three scientists that assessed both scientific and technical merits of this interesting contribution. As you will recognize from the enclosed comments, ref#1 advices to broaden the scope of the study addressing further Trf1's recruitment/possible Reb1 competition by further experimentation. Together with necessary clarifications in response to the rather explicit comments of our referees, we would be delighted to receive a revised version for final assessment. This might also present an opportunity to integrate the accepted study on Ydr026c you mentioned in recent communication into your discussion.

The Reb1-homologue Ydr026c/Nsi1 is required for efficient RNA polymerase I termination in yeast.

Several DNA cis-elements and trans-acting factors were described to be involved in transcription termination and to release the elongating RNA polymerases from their templates. Different models for the molecular mechanism of transcription termination have been suggested for eukaryotic RNA polymerase I (Pol I) from results of in vitro and in vivo experiments. To analyse the molecular requirement...

Co-transcriptional RNA cleavage provides a failsafe termination mechanism for yeast RNA polymerase I

Ribosomal RNA, transcribed by RNA polymerase (Pol) I, accounts for most cellular RNA. Since Pol I transcribes rDNA repeats with high processivity and polymerase density, transcription termination is a critical process. Early in vitro studies proposed polymerase pausing by Reb1 and transcript release at the T-rich element T1 determined transcription termination. However recent in vivo studies re...

The fission yeast rDNA-binding protein Reb1 regulates G1 phase under nutritional stress.

Yeast Reb1 and its mammalian ortholog TTF1 are conserved Myb-type DNA-binding proteins that bind to specific sites near the 3'-end of rRNA genes (rDNA). Here, they participate in the termination of transcription driven by RNA polymerase I and block DNA replication forks approaching in the opposite direction. We found that Schizosaccharomyces pombe Reb1 also upregulates transcription of the ste9...

Nsi1 plays a significant role in the silencing of ribosomal DNA in Saccharomyces cerevisiae

In eukaryotic cells, ribosomal DNA (rDNA) forms the basis of the nucleolus. In Saccharomyces cerevisiae, 100-200 copies of a 9.1-kb rDNA repeat exist as a tandem array on chromosome XII. The stability of this highly repetitive array is maintained through silencing. However, the precise mechanisms that regulate rDNA silencing are poorly understood. Here, we report that S. cerevisiae Ydr026c, whi...

Functional architecture of the Reb1-Ter complex of Schizosaccharomyces pombe.

Reb1 ofSchizosaccharomyces pomberepresents a family of multifunctional proteins that bind to specific terminator sites (Ter) and cause polar termination of transcription catalyzed by RNA polymerase I (pol I) and arrest of replication forks approaching the Ter sites from the opposite direction. However, it remains to be investigated whether the same mechanism causes arrest of both DNA transactio...
